Honnouji Academy is ruled over by students clad in special outfits. Student Council President Satsuki Kiryuin, along with the Honnouji Academy Elite Four, have the academy under their absolute control.

Ryuko has taken up residence with Mako's family, the Mankanshokus. Mako is ganged up on by the Tennis Club while on the way to school.

Satsuki Kiryuin has been using Life Fibers in her plot to conquer all of the nation's academies. To fight back, Ryuko's father, created a Kamui, a uniform woven entirely of Life Fibers.

It is Honnouji Academy No-Late Day, held each semester by the Disciplinary Committee, and everyone is in a tizzy. There are countless traps on the route to school, and if a student is late they are summarily expelled.

Ryuko is defeating Honnouji Academy's club presidents at a steady pace when Tsumugu Kinagase appears before her. Kinagase wields a machine gun that neutralizes the abilities of Goku Uniforms. He can hold off even Two-Star club presidents.

Uzu Sanageyama, speaks directly with Satsuki and is given permission to face Ryuko in battle. Sanageyama's dynamic visual acuity allows him to anticipate his opponent's every move and get the drop on them.
